Cooling Raspberry Pi Zero
I am currently using a Raspberry Pi Zero as a Pi-Hole. When running "vcgencmd measure_temp" in the command line, it shows that the Raspberry Pi Zero Temp = 54.1C. I am using an Aluminium heat sink. I also have spare copper heat sinks. Would a Copper Heat Sink provide better cooling?!

Nothing wrong with 54C. You're still a very long way from having any kind of temperature related issues.

There's no need for a heatsink of any flavour on a Zero. Regarding copper, it won't do any better than aluminium.
Though it has a higher thermal conductivity, it has a lower specific heat capacity. A combination of the two would work marginally better.
But as others have said, a Pi Zero doesn't need a heatsink at all and would be a waste of raw materials.

Only true for same weight of heatsinks, for two dimensionally equal heatsinks copper will be better than aluminium both in the short term (heat soak) and in the long term.

No need for cooling on a Pi Zero in normal ambient temperatures.
I put one at the top of an antenna mast and the heat from direct sunlight in the summer did for it.

Now there is a flirc case for the Raspberry PI zero. It might be a worthwhile option.

I got it from amazon about 2 years ago, got two versions of it (GPIO covered and GPIO exposed).
sad to say its no longer available.....
