Are you using the legacy driver or one of the KMS drivers?
Omxplayer won't run on Raspbian 10-Buster when you use the fkms or kms driver, i.e. when your /boot/config.txt file has in it this line:
Remove it and omxplayer works again.dtoverlay=vc4-kms-v3d
It should, but there's a minor bug in detecting the fkms or kms overlay.
