Yes, that card should be fine. Bit of an overkill, since the Pi does not support the U3 high speed writing, but it will work, and it still should be very fast. If you can find a Samsung EVO+ U1 micro SD card, it should be cheaper, and should be just as fast (if not faster) on a Pi.

Which OS are you running? If it's Raspbian Jessie Lite then that file won't exist, because it's part of the GUI configuration, and there is no GUI/desktop installed in Lite.

Sure. Any Pi will run Jessie Lite. There are no restrictions or minimum requirements. We've got Lite on our Zero (on a 4GB SD card, since Lite is so small).
