That depends what version you're upgrading from. Once you've upgraded to the more recent version, you'll not need a reboot.

I have tried updating one of our Pis twice using curl -sL https://raw.github.com/wireload/screenly-ose/master/misc/upgrade.sh | bash however both installs have failed both times at the final stage with a fatal error. Another update: We've now released another image that fixes an issue with the scroll bars. If you've already installed the existing image (or upgraded), just run the installer again.

This is true. The current disk image for Screenly OSE does not work with the Raspberry Pi 3. This is something that we are working on.
