Using /dev/sda to specify the boot drive is a bad idea. If you have more than one USB drive connected there is no guarantee the boot drive will be assigned the sda device. Using PARTUUID is the recommended (and more foolproof) way. You can find PARTUUID with this command. ls -l /dev/disk/by-partuui...

Setting a partitition as bootable (MBR) is a BIOS thing, the RPi doesn't have a BIOS, so it's 100% irrelevant.
